Description

COPS2 encodes subunit 2 of the COP9 signalosome (CSN), a multiprotein complex that regulates the ubiquitin-proteasome pathway by controlling the activity of cullin-RING E3 ubiquitin ligases. CSN2 is essential for deneddylation of cullins, thereby modulating protein degradation, cell cycle progression, and signal transduction. It also functions as a transcriptional corepressor via interaction with nuclear receptors and other transcription factors.

Protein Summary

COPS2 (CSN2) is a 443-amino acid protein that forms an integral part of the COP9 signalosome complex. It contains a PCI domain that mediates complex assembly. CSN2 is required for the deneddylation of cullin proteins, which inactivates cullin-RING E3 ligases and prevents premature substrate ubiquitination. Additionally, CSN2 acts as a transcriptional corepressor by recruiting histone deacetylases to nuclear receptors such as thyroid hormone receptor and retinoic acid receptor. Its dual role in proteolysis and transcription links it to cell growth, differentiation, and cancer.
