Description

CDK19 (Cyclin Dependent Kinase 19) is a member of the cyclin-dependent kinase family. It is a component of the Mediator complex, which is essential for transcriptional regulation. CDK19 phosphorylates the C-terminal domain (CTD) of RNA polymerase II, thereby influencing transcription initiation and elongation. It also plays a role in RNA processing and cell cycle regulation. Mutations and altered expression of CDK19 have been associated with various cancers and developmental disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Altered expression and mutations in CDK19 can disrupt transcriptional regulation, promoting uncontrolled cell proliferation. COSMIC, ClinVar
Developmental disorders Germline mutations in CDK19 may affect neurodevelopment and other developmental processes. OMIM, ClinVar

Mutation functional classification

Loss of Function (LOF)

Loss-of-function mutations in CDK19 are rare but may impair Mediator complex function and transcriptional regulation.

Gain of Function (GOF)

Amplification or overexpression of CDK19 may act as a gain-of-function, promoting oncogenic transcription.

Dominant Negative (DN)

Dominant-negative effects have not been clearly established for CDK19 mutations.

Pathways

• Mediator complex
• RNA polymerase II transcription
• Cell cycle

Protein Summary

CDK19 is a serine/threonine protein kinase that phosphorylates the C-terminal domain of RNA polymerase II. It is a core component of the Mediator complex, which bridges transcription factors and RNA polymerase II. CDK19 activity is regulated by cyclin binding and phosphorylation. The protein is involved in transcription initiation, elongation, and RNA processing. Its dysregulation is linked to cancer and developmental disorders.
