Description

CDH26 (Cadherin 26) encodes a non-classical cadherin that lacks the typical HAV cell adhesion recognition motif. It is involved in calcium-dependent cell-cell adhesion, immune modulation, and epithelial barrier function. CDH26 is expressed in the respiratory and gastrointestinal tracts and has been implicated in asthma, allergic inflammation, and various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Asthma CDH26 variants may alter airway epithelial integrity and immune response, contributing to asthma susceptibility. ClinVar, OMIM
Allergic rhinitis Polymorphisms in CDH26 are associated with increased risk of allergic rhinitis in genome-wide studies. NCBI Gene, OMIM
Colorectal cancer Reduced CDH26 expression is observed in colorectal tumors, suggesting a tumor-suppressive role through loss of cell adhesion. COSMIC, NCBI Gene
Ovarian cancer CDH26 hypermethylation and downregulation are linked to ovarian cancer progression and metastasis. COSMIC, PubMed
Inflammatory bowel disease CDH26 expression is altered in intestinal epithelium of IBD patients, potentially affecting barrier function. NCBI Gene

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations (e.g., c.2345_2346insA, c.3010C>T) are predicted to cause loss of function through protein truncation or nonsense-mediated decay.

Gain of Function (GOF)

No gain-of-function mutations have been reported for CDH26.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for CDH26.

Pathways

Cell adhesion molecules (CAMs) – KEGG hsa04514
Adherens junction – KEGG hsa04520
Wnt signaling pathway (indirect via β-catenin interaction)

Protein Summary

Cadherin 26 is a 1004-amino acid transmembrane glycoprotein belonging to the cadherin superfamily. It contains five extracellular cadherin repeats, a transmembrane domain, and a cytoplasmic tail. Unlike classical cadherins, CDH26 lacks the HAV motif and may mediate calcium-dependent adhesion through alternative mechanisms. The protein is expressed in epithelial tissues, particularly in the lung and gastrointestinal tract, and plays roles in maintaining barrier integrity and immune homeostasis. Altered expression or mutation of CDH26 is associated with asthma, allergic diseases, and epithelial cancers.
