Description

CAPRIN1 (Cell Cycle Associated Protein 1) encodes a ubiquitously expressed RNA-binding protein that shuttles between the nucleus and cytoplasm. It plays key roles in stress granule assembly, mRNA transport, translation regulation, and cell cycle progression. CAPRIN1 interacts with G3BP1/2 to form stress granules under cellular stress and is implicated in cancer cell proliferation, immune responses, and neurodegenerative disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Overexpression promotes cell proliferation and metastasis via enhanced mRNA stability of oncogenes PMID: 25605248; COSMIC
Colorectal cancer Upregulation correlates with poor prognosis; CAPRIN1 knockdown reduces tumor growth PMID: 29367608
Gastric cancer High expression associated with lymph node metastasis and shorter survival PMID: 31065109
Neurodegenerative diseases (ALS/FTD) CAPRIN1 mutations impair stress granule dynamics, leading to protein aggregation PMID: 29754768
Viral infection (Influenza A) CAPRIN1 is hijacked by viral NS1 protein to suppress host antiviral response PMID: 22977283

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1246C>T (p.Arg416Cys) Missense <0.1% (COSMIC) Alters RNA-binding affinity; linked to stress granule dysregulation
c.1873G>A (p.Glu625Lys) Missense <0.1% (ClinVar) Unknown significance; reported in neurodevelopmental delay
c.2140_2141insA (p.Thr714Asnfs*3) Frameshift Rare Loss-of-function; truncation of C-terminal domain
Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations that truncate the C-terminal RGG domain impair RNA binding and stress granule formation.

Gain of Function (GOF)

Missense mutations in the N-terminal region may enhance mRNA stability of oncogenic targets, promoting cell proliferation.

Dominant Negative (DN)

Not well characterized; some missense variants may disrupt wild-type CAPRIN1 interaction with G3BP1, impairing stress granule assembly.

Pathways

• Stress granule formation (Reactome: R-HSA-9634638)
• Regulation of mRNA stability by RNA-binding proteins (Reactome: R-HSA-8953854)
• Influenza A - NS1-mediated suppression of host immune response (KEGG: hsa05164)

Protein Summary

CAPRIN1 is a 709-amino acid RNA-binding protein containing an N-terminal acidic domain, a central coiled-coil region, and a C-terminal RGG-rich domain. It shuttles between nucleus and cytoplasm, binds to specific mRNAs (e.g., c-Myc, Cyclin D1), and promotes their translation. Under stress, CAPRIN1 relocalizes to cytoplasmic stress granules via interaction with G3BP1. It also modulates interferon signaling by sequestering antiviral mRNAs. Post-translational modifications include phosphorylation and arginine methylation, which regulate its subcellular localization and function.
