Description
CAPN15 (calpain 15) is a protein-coding gene that encodes a member of the calpain family of intracellular cysteine proteases. The protein contains a calpain catalytic domain and a C-terminal domain homologous to the Drosophila small optic lobes (SOL) protein, suggesting a role in neuronal development and function. CAPN15 is involved in proteolytic processing of target proteins, regulating cellular processes such as cell cycle, differentiation, and apoptosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Colorectal cancer | Altered expression and activity of CAPN15 may contribute to tumor progression through dysregulation of proteolytic signaling. | COSMIC; PMID: 23549388 |
| Breast cancer | CAPN15 mutations and expression changes have been observed in breast cancer samples, potentially affecting cell adhesion and migration. | COSMIC; PMID: 25691885 |
| Lung cancer | Somatic mutations in CAPN15 have been identified in lung adenocarcinoma, with possible impact on cell survival pathways. | COSMIC; PMID: 22980975 |

Protein Summary
Calpain 15 (CAPN15) is a 729-amino acid calcium-dependent cysteine protease. It contains a catalytic domain typical of calpains and a unique C-terminal SOLH domain. The protein is expressed in multiple tissues, with highest levels in brain and testis. It is involved in limited proteolysis of substrates, influencing cell signaling, cytoskeletal remodeling, and development. Mutations in CAPN15 are rare but have been reported in various cancers.
