Description

CADM3 (Cell Adhesion Molecule 3) encodes a member of the immunoglobulin superfamily. The protein is a transmembrane cell adhesion molecule that mediates homophilic and heterophilic cell-cell interactions. It is involved in neuronal synapse organization, tumor suppression, and epithelial integrity. CADM3 is also known as TSLC1-like, IgSF4C, NECL1, and SynCAM3.

Loss of Function (LOF)

Nonsense mutations (e.g., p.Arg412*) lead to truncated protein and loss of adhesion function.

Gain of Function (GOF)

No gain-of-function mutations reported in CADM3.

Dominant Negative (DN)

No dominant-negative mutations described for CADM3.

Protein Summary

CADM3 is a 445-amino acid transmembrane glycoprotein with three extracellular immunoglobulin-like domains (one V-type, two C2-type) and a short cytoplasmic tail. It mediates homophilic and heterophilic adhesion with other CADM family members. The protein is highly expressed in brain and testis, and plays roles in synapse formation, neurite outgrowth, and tumor suppression. Loss of CADM3 expression is observed in several cancers.
