Description

CAB39L (Calcium Binding Protein 39 Like) encodes a scaffold protein that forms a heterotrimeric complex with STK11 (LKB1) and STRAD (STE20-related kinase). This complex is essential for LKB1 kinase activity, which regulates cell polarity, energy metabolism, and tumor suppression. CAB39L is widely expressed and acts as a cofactor for LKB1-mediated phosphorylation of AMPK and other AMPK-related kinases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (general) Loss of CAB39L expression or complex disruption impairs LKB1 tumor suppressor function, promoting cell proliferation and metastasis. PMID: 19029980
Peutz-Jeghers Syndrome (related) CAB39L mutations are not directly causative but may modify LKB1 pathway activity in hamartomatous polyposis. OMIM: 612175

Protein Summary

CAB39L (MO25 beta) is a 341-amino acid scaffold protein that binds both LKB1 and STRAD, stabilizing the active LKB1-STRAD-MO25 complex. It does not possess intrinsic enzymatic activity but is critical for LKB1 kinase activation. The protein contains a conserved calcium-binding EF-hand domain, though calcium binding is not required for complex formation. CAB39L is ubiquitously expressed with highest levels in testis and brain.
