Description

BRMS1 (Breast Cancer Metastasis Suppressor 1) is a protein-coding gene that encodes a member of the mSin3A histone deacetylase (HDAC) complex. The protein functions as a metastasis suppressor, inhibiting the spread of cancer cells from primary tumors to distant sites without affecting primary tumor growth. It is involved in transcriptional regulation, chromatin remodeling, and suppression of NF-kappaB signaling. BRMS1 is downregulated in several metastatic cancers, and its re-expression reduces metastatic potential in experimental models.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast Cancer Loss of BRMS1 expression correlates with increased metastasis; re-expression suppresses metastasis via HDAC complex and NF-κB inhibition PMID: 12676963, PMID: 18245496
Melanoma BRMS1 downregulation promotes metastasis; restoration reduces metastatic burden in xenograft models PMID: 19029987
Non-Small Cell Lung Cancer Reduced BRMS1 expression associated with lymph node metastasis and poor prognosis PMID: 21502317
Ovarian Cancer BRMS1 loss linked to peritoneal metastasis and shorter survival PMID: 20629107

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.256C>T (p.Arg86Trp) Missense Rare Unknown functional impact

c.1A>G (p.Met1Val) Start loss Rare Likely loss of function
Mutation functional classification

Loss of Function (LOF)

Start loss mutations (e.g., p.Met1Val) and deletions are predicted to cause loss of BRMS1 protein function, reducing metastasis suppression.

Gain of Function (GOF)

No gain-of-function mutations have been reported for BRMS1.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for BRMS1.

Pathways

• HDAC complex pathway (mSin3A complex)
• NF-kappaB signaling pathway (negative regulation)

Protein Summary

The BRMS1 protein is a 246-amino-acid component of the mSin3A histone deacetylase (HDAC) complex. It localizes to the nucleus and functions as a transcriptional corepressor. BRMS1 suppresses metastasis by modulating chromatin structure, repressing pro-metastatic genes, and inhibiting NF-κB activity. It does not affect primary tumor growth but specifically blocks the dissemination of cancer cells to distant organs. The protein contains two coiled-coil domains and a nuclear localization signal.
