Description

ALOX15B encodes a member of the lipoxygenase family that converts arachidonic acid to 15(S)-hydroperoxyeicosatetraenoic acid (15(S)-HPETE). The enzyme is involved in lipid peroxidation, inflammation, and cellular signaling. It is predominantly expressed in epithelial tissues and has been implicated in prostate cancer and other malignancies.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate Cancer Reduced expression of ALOX15B is associated with tumor progression; loss of 15-LOX-2 activity may promote carcinogenesis through altered eicosanoid signaling. PMID: 11526490, PMID: 12619171
Atherosclerosis ALOX15B contributes to oxidative modification of low-density lipoproteins (LDL) in the arterial wall, promoting foam cell formation. PMID: 10488063
Inflammatory Bowel Disease Altered ALOX15B expression in colonic mucosa may modulate inflammation via production of anti-inflammatory lipid mediators. PMID: 16959936

Protein Summary

ALOX15B encodes a 676-amino acid protein (15-lipoxygenase-2) that contains a non-heme iron catalytic domain. The enzyme specifically oxygenates arachidonic acid at carbon 15 to produce 15(S)-HPETE, which is further metabolized to anti-inflammatory lipoxins and resolvins. The protein is localized to the cytoplasm and plasma membrane. Structural studies reveal a two-domain architecture: an N-terminal β-barrel domain and a C-terminal catalytic domain.
