Description
ADAMTS15 encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) family of zinc-dependent metalloproteinases. The encoded preproprotein is proteolytically processed to generate the mature enzyme, which functions in the degradation of proteoglycans such as aggrecan and versican. ADAMTS15 is involved in extracellular matrix remodeling, cell migration, and has been implicated in cancer progression and osteoarthritis.

Protein Summary
ADAMTS15 is a secreted metalloproteinase composed of a signal peptide, a prodomain, a catalytic domain with a zinc-binding motif, a disintegrin-like domain, a central thrombospondin type 1 repeat, a cysteine-rich domain, and a spacer region. The enzyme cleaves aggrecan at the Glu373-Ala374 bond and versican at the Glu441-Ala442 bond. Its activity is regulated by tissue inhibitors of metalloproteinases (TIMPs). ADAMTS15 is widely expressed in adult tissues, with highest levels in testis and colon. Reduced expression is observed in several cancers, suggesting a tumor-suppressive role.
