Description

TEAD1 (TEA domain transcription factor 1) encodes a transcription factor that binds to the MCAT element in muscle-specific genes and is a key downstream effector of the Hippo signaling pathway. It regulates cell proliferation, differentiation, and organ size by interacting with co-activators such as YAP1 and WWTR1 (TAZ). TEAD1 is essential for embryonic development, particularly in heart, muscle, and neural tube formation.

Mutation functional classification

Loss of Function (LOF)

Mutations that disrupt DNA binding or co-activator interaction, leading to reduced transcriptional activity (e.g., p.Thr166Ile).

Gain of Function (GOF)

Amplification or overexpression of TEAD1, often with YAP1, promoting oncogenic transcription.

Dominant Negative (DN)

Not well documented; some missense variants may interfere with wild-type TEAD1 function.

Protein Summary

TEAD1 is a 426-amino acid transcription factor containing a conserved TEA/ATTS DNA-binding domain. It forms complexes with YAP1/TAZ to regulate gene expression involved in cell growth, apoptosis, and differentiation. The protein is predominantly nuclear and is widely expressed in adult tissues, with highest levels in heart and skeletal muscle.
