Description

LATS2 (Large Tumor Suppressor Kinase 2) is a serine/threonine protein kinase that functions as a core component of the Hippo signaling pathway. It phosphorylates and inactivates YAP1 and WWTR1/TAZ transcriptional coactivators, thereby regulating cell proliferation, apoptosis, and organ size. LATS2 also plays a role in mitotic progression, centrosome duplication, and DNA damage response. Loss of LATS2 function is implicated in various cancers, including breast, lung, and prostate cancer.

Protein Summary

LATS2 encodes a 1088-amino acid serine/threonine kinase with an N-terminal UBA domain, a central kinase domain, and a C-terminal regulatory region. It is activated by MST1/2-mediated phosphorylation and forms a complex with MOB1 to phosphorylate YAP1 and TAZ, leading to their cytoplasmic retention and degradation. LATS2 also localizes to centrosomes and mitotic spindles, where it regulates centrosome duplication and mitotic fidelity. The protein is widely expressed, with highest levels in testis and heart.
