Description

ADIPOR2 (Adiponectin Receptor 2) is a protein-coding gene that encodes a receptor for adiponectin, a hormone secreted by adipose tissue involved in glucose and lipid metabolism. The receptor mediates increased AMPK and PPAR-alpha ligand activities, leading to fatty acid oxidation and glucose uptake. ADIPOR2 is widely expressed and plays a role in insulin sensitivity, inflammation, and energy homeostasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Type 2 Diabetes Reduced ADIPOR2 expression impairs adiponectin signaling, decreasing insulin sensitivity and glucose uptake. ClinVar, NCBI
Obesity Altered ADIPOR2 levels correlate with adiposity and metabolic dysfunction. NCBI, OMIM
Metabolic Syndrome Dysregulation of ADIPOR2 contributes to insulin resistance, dyslipidemia, and hypertension. NCBI
Non-alcoholic Fatty Liver Disease (NAFLD) Reduced hepatic ADIPOR2 expression is associated with steatosis and inflammation. NCBI
Cardiovascular Disease Impaired adiponectin signaling via ADIPOR2 may promote atherosclerosis. NCBI

Protein Summary

Adiponectin receptor 2 (ADIPOR2) is a 375-amino acid transmembrane protein with seven membrane-spanning domains, structurally distinct from classical GPCRs. It binds adiponectin and mediates downstream activation of AMPK and PPAR-alpha, promoting fatty acid oxidation, glucose uptake, and anti-inflammatory effects. ADIPOR2 is ubiquitously expressed, with highest levels in skeletal muscle, liver, and adipose tissue.
