Description
ADGRG3 (Adhesion G Protein-Coupled Receptor G3), also known as GPR97, is a member of the adhesion G protein-coupled receptor (GPCR) family. It is characterized by a large extracellular N-terminal domain containing adhesion-like motifs. The receptor is involved in cell adhesion, migration, and signaling, and is expressed in various tissues including the brain, immune cells, and reproductive organs. ADGRG3 has been implicated in immune regulation and certain cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast cancer | Overexpression of ADGRG3 may promote tumor cell migration and invasion via GPCR signaling pathways. | COSMIC; PMID: 25691885 |
| Prostate cancer | ADGRG3 expression is altered in prostate cancer tissues, potentially affecting androgen receptor signaling. | COSMIC; PMID: 27387371 |
| Inflammatory bowel disease | ADGRG3 variants may influence immune cell adhesion and gut inflammation. | ClinVar; PMID: 28067908 |

Protein Summary
ADGRG3 encodes a 7-transmembrane adhesion GPCR with a large extracellular domain containing a GPS (GPCR proteolysis site) motif. The protein undergoes autoproteolytic cleavage at the GPS site, resulting in an N-terminal fragment (NTF) and a C-terminal fragment (CTF) that remain non-covalently associated. ADGRG3 is involved in cell-cell and cell-matrix adhesion, and its signaling modulates cell migration and immune responses. Expression is enriched in brain, spleen, and reproductive tissues.
