Protein Summary

ADGRA3 is a 1,200-amino acid adhesion GPCR with a large extracellular region containing a GPS (GPCR proteolysis site) domain, a seven-transmembrane domain, and a cytoplasmic tail. It undergoes autoproteolytic cleavage at the GPS motif, producing an N-terminal fragment (NTF) and a C-terminal fragment (CTF) that remain non-covalently associated. The protein mediates cell adhesion and may activate G proteins (e.g., Gα12/13) to modulate Rho signaling. Expression is highest in testis and brain, with aberrant expression in several cancers.
